If your device does not have location service enabled, this problem may occur. Please follow the steps below to check if location service is enabled: 
Scroll down the control panel on the device, then find the "Position" icon. If the button's color is blue, it means you have enabled it. If not, click on the button to activate the positioning service.

Yes, if you change the location using iGPS Spoofer, the virtual location will be retained. Unless you turn off your iPhone and then restart it. Or on AnyiGPS Spoofer, you can "Reset Location" to restore the actual location.
